A practical reference on Ipamorelin: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.
This page was last updated on 2025-09-13 and is reviewed periodically as new material appears.
Selectivity distinguishes ipamorelin from first-generation secretagogues such as GHRP-6. At doses that reliably raise growth hormone, it shows little stimulation of adrenocorticotropic hormone or cortisol release in animal models, and it does not markedly raise prolactin or appetite. Binding at GHS-R1a on pituitary somatotrophs triggers calcium influx and pulsatile growth hormone secretion. Because the compound mimics the natural ghrelin signal, the release pattern tends to follow the body's own rhythm rather than producing a sustained elevation.
Most published work on ipamorelin comes from rodent studies and small early-phase human trials. Subcutaneous and intravenous routes have been used, while oral delivery is limited by poor absorption and rapid breakdown in the gut. The reported plasma half-life is short, on the order of two hours, and varies with species and assay method. Whether chronic use produces meaningful clinical benefit remains unresolved, and long-term safety data in humans are sparse. No major regulatory agency has approved the compound as a therapeutic drug.

Ipamorelin is a synthetic pentapeptide that acts on the growth hormone secretagogue receptor, also known as the ghrelin receptor. Its sequence contains five amino acid residues, including a non-natural residue that increases stability against enzymatic breakdown. The compound was developed in the 1990s as part of research into small peptides that stimulate pituitary hormone release. Unlike larger protein hormones, it can be produced by solid-phase peptide synthesis and characterized by standard analytical methods.
At the receptor level, ipamorelin binds GHS-R1a and triggers signaling through Gq-coupled pathways. Activation leads to calcium release and downstream effects in pituitary somatotroph cells. These events promote the release of growth hormone into circulation. The response depends on the presence of the receptor and on the physiological state of the animal or tissue studied. Because the receptor is also found in other tissues, effects beyond the pituitary have been examined in laboratory models, though the extent of those effects remains an area of ongoing study.
One distinguishing feature reported in animal studies is selectivity. Ipamorelin stimulated growth hormone release with limited elevation of adrenocorticotropic hormone or cortisol compared with earlier secretagogues such as GHRP-6. This pattern has been described as more selective for the growth hormone axis. The finding comes mainly from preclinical work, and the degree to which it holds across species and doses is not fully settled. Reports also describe effects on gastric motility in animal models, suggesting activity outside the pituitary, though the clinical relevance of this observation is uncertain.
Ipamorelin is a synthetic pentapeptide classified as a growth hormone secretagogue. Its sequence, Aib-His-D-2-Nal-D-Phe-Lys-NH2, combines three non-proteinogenic residues with a C-terminal amide. The N-terminal aminoisobutyric acid unit and the two aromatic D-amino acids distinguish it from peptides assembled only from standard L-amino acids. Its formula is C38H49N9O5, corresponding to an average mass near 711.9 Da. At neutral pH the molecule carries a net positive charge, a property that shapes its behaviour in chromatographic and electrophoretic systems.
The compound was developed at Novo Nordisk during the 1990s as part of a programme seeking secretagogues with improved selectivity. It was described in the peer-reviewed literature in 1998 alongside related pentapeptides from the same series. Investigators advanced it because it raised growth hormone output in animal models while leaving other pituitary hormones comparatively unaffected. The development code NNC 26-0161 appears in earlier reports, and ipamorelin later became the common designation in published work.
During atrial systole, blood flows from the atria to the ventricles down the pressure gradient. Chordae tendineae are relaxed because the atrioventricular (AV) valves are forced open. When the ventricles of the heart contract in ventricular systole, the increased blood pressures in both chambers push the AV valves to close simultaneously, preventing the backflow of blood into the atria. Since the blood pressure in the atria is much lower than that in the ventricles, the flaps attempt to evert to the low pressure regions. The chordae tendineae prevent this prolapse by becoming tense, which pulls on the flaps, holding them in a closed position.

=== RA 27/3 === Most of the modern Rubella vaccines (including the combination vaccine MMR) contain the RA 27/3 strain, which was developed by Stanley Plotkin and Leonard Hayflick at the Wistar Institute in Philadelphia. The vaccine was attenuated and prepared in the WI-38 normal human diploid cell strain which was developed by Hayflick and provided to Plotkin by him. In order to isolate the virus, instead of taking swab samples from the throats of infected patients, which could have been contaminated with other resident viruses, Plotkin decided to utilize aborted fetuses provided by the department of Obstetrics and Gynecology of the Hospital of the University of Pennsylvania. At the time, abortion was illegal in most of the United States (including Pennsylvania), but doctors were allowed to perform "therapeutic abortions" when the life of the woman was in danger. Some started to perform them also on women infected with rubella. Several dozens of aborted fetuses were collected and studied by Plotkin. The kidney tissue from fetus 27 produced the strain that was used to develop the attenuated rubella vaccine. The name RA 27/3 refers to "Rubella Abortus", 27th fetus, 3rd organ to be harvested (the kidney). The vaccine was first approved by the UK in 1970. The strain became the preferred vaccine used by pharmaceutical companies over the HPV-77, due to several considerations, including its higher immunogenicity; Merck made it its mainstay rubella vaccine in 1979. Parkman and his team did not monetize their patents, wanting the vaccine to be freely available.
=== Direct DNA template hypothesis === Soviet-American physicist George Gamow was the first to come up with a coherent scheme for protein synthesis from DNA. Based on the Watson-Crick model, he envisaged that the DNA itself is a direct template for protein synthesis. Assuming that the four bases of DNA could produce 20 different combinations as triplets, he suggested that the different amino acids must correspond to a twenty-letter alphabet of the nucleotide sequence. In such configuration, the DNA directly produces proteins from the free molecules of amino acids. In the 13 February 1954 issue of Nature he explained:It seems to me that such translation procedure can be easily established by considering the 'key-and-lock' relation between various amino-acids, and the rhomb-shaped 'holes' formed by various nucleotides in the deoxyribonucleic acid chain... One can speculate that free amino-acids from the surrounding medium get caught into the 'holes' of deoxyribonucleic acid molecules, and thus unite into the corresponding peptide chains.Watson, then at the California Institute of Technology in Massachusetts, also thought up the issue. Unlike Gamow, he realised the possible importance of RNA as an intermediate stage in protein synthesis. He initially imagined that DNA was first converted by chemical reaction to RNA, but then changed his view to DNA as a template for RNA synthesis. This latter model, which he called "not ugly" was starting to become more convincing.

Although 54.0% of the population lived in urban areas in 2011, this percentage has been declining since 1996. Counties with over 2⁄3 urban population are Hunedoara, Brașov and Constanța, while those with less than a third are Dâmbovița (30.06%) and Giurgiu and Teleorman. Bucharest is the capital and the largest city in Romania, with a population of over 1.7 million in 2021. Its larger urban zone has a population of almost 2.2 million, which are planned to be included into a metropolitan area up to 20 times the area of the city proper. Another 17 cities have a population of over 100,000, with Cluj-Napoca, Iași, Constanța and Timișoara having more than 250,000 inhabitants, and Craiova, Brașov and Galați with over 200,000 inhabitants. Metropolitan areas have been constituted for most of these cities.
=== Sarcomere assembly === In addition to sarcomere activity, it has been shown that myomesin also plays a role in the assembly of the sarcomere. In order for myomesin to be implemented into the sarcomere, myosin and titin must be present, indicating that myomesin is the last component to be added during assembly of the lattice. It is believed that this postponed addition is due to the role of myomesin to act as an "integrity check" to ensure the sarcomere has been formed correctly and monitor its integrity. This is extremely important as if even one piece of the M-line is missing, the A-band of the sarcomere will collapse and the muscle will be paralyzed.
During the mid-19th century, one of the most remarkable painters was Ramón Torres Méndez, who produced a series of good quality paintings depicting the people and their customs of different Colombian regions. Also noteworthy in the 19th century were Andrés de Santa María, Pedro José Figueroa, Epifanio Garay, Mercedes Delgado Mallarino, José María Espinosa, Ricardo Acevedo Bernal, between many others. More recently, Colombian artists Pedro Nel Gómez and Santiago Martínez Delgado started the Colombian Murial Movement in the 1940s, featuring the neoclassical features of Art Deco. Since the 1950s, the Colombian art started to have a distinctive point of view, reinventing traditional elements under the concepts of the 20th century. Examples of this are the Greiff portraits by Ignacio Gómez Jaramillo, showing what the Colombian art could do with the new techniques applied to typical Colombian themes. Carlos Correa, with his paradigmatic "Naturaleza muerta en silencio" (silent dead nature), combines geometrical abstraction and cubism. Alejandro Obregón is often considered as the father of modern Colombian painting, and one of the most influential artist in this period, due to his originality, the painting of Colombian landscapes with symbolic and expressionist use of animals, (specially the Andean condor). Fernando Botero, Omar Rayo, Enrique Grau, Édgar Negret, David Manzur, Rodrigo Arenas Betancourt, Oscar Murillo, Doris Salcedo and Oscar Muñoz are some of the Colombian artists featured at the international level.

The peptide is Aib-His-D-2-Nal-D-Phe-Lys-NH2. Two of its residues are non-natural, which slows enzymatic degradation. The C-terminal amide is common among bioactive peptides.
It binds the same ghrelin receptor but with greater selectivity in functional assays. Preclinical work reports less cortisol and prolactin stimulation at growth-hormone-releasing doses. Those differences are relative, not absolute, and depend on dose and model.
No major regulatory authority has approved it for human therapeutic use. It is sold as a research chemical for laboratory investigation. Clinical status varies by country and is subject to change.
