At the molecular level, ipamorelin acts as an agonist at the growth hormone secretagogue receptor, also called the ghrelin receptor or GHS-R1a. Binding to this receptor on pituitary somatotroph cells triggers a signaling cascade that leads to growth hormone release. The effect is mediated through phospholipase C and calcium mobilization rather than through the cyclic AMP pathway used by growth hormone releasing hormone. The two pathways are complementary, and combined stimulation produces a larger response than either alone.
Compared with other secretagogues such as GHRP-2, GHRP-6, and hexarelin, ipamorelin is described as more selective. Published animal work reports little or no increase in adrenocorticotropic hormone, cortisol, or prolactin at doses that release growth hormone. This selectivity is the property most often cited in the research literature. Whether the same profile holds across species and dosing schedules remains an open question, since human data are limited and come largely from small studies.
Ipamorelin is a synthetic pentapeptide that belongs to the growth hormone secretagogue class. It acts as an agonist at the ghrelin receptor, also called the growth hormone secretagogue receptor type 1a. The compound was designed in the 1990s during a search for agents that release growth hormone with fewer off-target hormonal effects than earlier secretagogues. It appears in the research literature under several sequence-based names. Material supplied for laboratory work is normally a lyophilized solid, and it is not marketed as an approved therapeutic in major jurisdictions.
The molecule contains five residues, including alpha-aminoisobutyric acid, D-2-naphthylalanine, and D-phenylalanine, and it ends in a lysine amide. Non-natural and D-configured residues make the chain less susceptible to common peptidases, which helps explain its resistance to rapid breakdown. Its molecular formula is C38H49N9O5, corresponding to a free-base mass near 711.9 daltons. The C-terminal amide removes a negative charge and is a recurring feature in receptor-active peptides of this family. These structural choices are usually discussed as the basis for its selectivity profile.
Published animal and early human work describes growth hormone release that is separated from comparable rises in adrenocorticotropic hormone and cortisol. Prolactin changes are reported as small in the same studies. Selectivity is attributed to binding at the ghrelin receptor and to the downstream signaling that follows, rather than to differences in how quickly the peptide is cleared. Authors commonly label the compound selective rather than potent, because the same mass produces a smaller growth hormone response than some older secretagogues tested in parallel. Whether that profile holds across species and routes of administration remains an open question.

Signal transduction begins when the peptide binds GHSR-1a on pituitary somatotrophs. The receptor couples to Gq/11 proteins, activating phospholipase C, which cleaves phosphatidylinositol bisphosphate into inositol trisphosphate and diacylglycerol. Inositol trisphosphate releases calcium from intracellular stores, and the resulting rise in cytosolic calcium drives growth hormone vesicle fusion. Concurrent Gs coupling and cyclic AMP elevation have also been reported, and the relative contribution of each arm to the overall secretory response is not fully settled.
Structural features distinguish the molecule from earlier secretagogues. An alpha-aminoisobutyric acid residue near the N-terminus and a D-naphthylalanine substitution increase receptor affinity, while C-terminal amidation improves resistance to exopeptidases. These modifications are associated with reduced stimulation of appetite and of the hypothalamic-pituitary-adrenal axis compared with hexarelin or growth hormone releasing peptide-6. Whether the same profile applies at every dose level studied is a matter of ongoing investigation rather than settled consensus.
=== Steam cracking === Steam cracking is the process for producing ethylene and other alkenes from aliphatic hydrocarbons. Depending on the feedstock used to produce the olefins, steam cracking can produce a benzene-rich liquid by-product called pyrolysis gasoline. Pyrolysis gasoline can be blended with other hydrocarbons as a gasoline additive, or routed through an extraction process to recover BTX aromatics (benzene, toluene and xylenes).
The hydrolysis of GTP allows for a large conformational change within EF-G, forcing the A/P tRNA to fully occupy the P site, the P/E tRNA to fully occupy the E site (and exit the ribosome complex), and the mRNA to shift three nucleotides down relative to the ribosome. The GDP-bound EF-G molecule then dissociates from the complex, leaving another free A-site where the elongation cycle can start again.
== Pathogenesis == The cells of adipose (fat) tissue synthesize hormones known as adipokines. In humans, dysfunction of adipose tissue, even in cases without obesity, has been associated with the development of insulin resistance, hypertension, systemic inflammation, and increased risk of blood clots (thrombosis). The inflammation produced by these hormones are thought to inflame adipose tissue, leading to the production of more adipokines and perpetuation of the cycle, and a constant low-level, pro-inflammatory state. Although it is suspected that a similar mechanism occurs in horses, further research is needed.
=== Classification methods === Classification methods use data to train a program (classifier) to distinguish positive examples of interacting protein/domain pairs with negative examples of non-interacting pairs. Popular classifiers used are Random Forest Decision (RFD) and Support Vector Machines. RFD produces results based on the domain composition of interacting and non-interacting protein pairs. When given a protein pair to classify, RFD first creates a representation of the protein pair in a vector. The vector contains all the domain types used to train RFD, and for each domain type the vector also contains a value of 0, 1, or 2. If the protein pair does not contain a certain domain, then the value for that domain is 0. If one of the proteins of the pair contains the domain, then the value is 1. If both proteins contain the domain, then the value is 2. Using training data, RFD constructs a decision forest, consisting of many decision trees. Each decision tree evaluates several domains, and based on the presence or absence of interactions in these domains, makes a decision as to if the protein pair interacts. The vector representation of the protein pair is evaluated by each tree to determine if they are an interacting pair or a non-interacting pair. The forest tallies up all the input from the trees to come up with a final decision. The strength of this method is that it does not assume that domains interact independent of each other. This makes it so that multiple domains in proteins can be used in the prediction.
== Collaborations == In 2013, La Colombe partnered with the Clinton Foundation to found the Haiti Coffee Academy. The organization's stated aim is to improve coffee growers’ livelihoods in Haiti through training programs for smallholder farmers, a demonstration farm and nursery, and efforts to strengthen the supply chain for both local and export markets for Haitian coffee. In 2016, La Colombe collaborated with Yards Brewing Company, also in the Philadelphia area, to produce a coffee stout to be served in local Shake Shack branches. Also in 2016, the company collaborated with the Icelandic brewer Einstök and again with Yards on Snorri's Awakening, a coffee pale ale. In 2021, the company collaborated with Whole Foods Market to open new stores in New York City and Austin.

Catabolism () is the set of metabolic pathways that breaks down molecules into smaller units that are either oxidized to release energy or used in other anabolic reactions. Catabolism breaks down large molecules (such as polysaccharides, lipids, nucleic acids, and proteins) into smaller units (such as monosaccharides, fatty acids, nucleotides, and amino acids, respectively). Catabolism is the breaking-down aspect of metabolism, whereas anabolism is the building-up aspect. Cells use the monomers released from breaking down polymers to either construct new polymer molecules or degrade the monomers further to simple waste products, releasing energy. Cellular wastes include lactic acid, acetic acid, carbon dioxide, ammonia, and urea. The formation of these wastes is usually an oxidation process involving a release of chemical free energy, some of which is lost as heat, but the rest of which is used to drive the synthesis of adenosine triphosphate (ATP). This molecule acts as a way for the cell to transfer the energy released by catabolism to the energy-requiring reactions that make up anabolism. Catabolism is a destructive metabolism and anabolism is a constructive metabolism. Catabolism, therefore, provides the chemical energy necessary for the maintenance and growth of cells. Examples of catabolic processes include glycolysis, the citric acid cycle, the breakdown of muscle protein in order to use amino acids as substrates for gluconeogenesis, the breakdown of fat in adipose tissue to fatty acids, and oxidative deamination of neurotransmitters by monoamine oxidase.

==== Role in cardiac hypertrophy and fibrosis ==== Intracrine Ang II has been implicated in the development of cardiac hypertrophy, a process characterized by the enlargement of cardiac myocytes in response to increased workload or stress. Experimental models have shown that overexpression of non-secreted Ang II in cardiac cells leads to rapid hypertrophy independent of extracellular Ang II signaling. This suggests that intracellular Ang II plays a direct role in cardiomyocyte growth and structural remodeling. Similarly, intracrine Ang II contributes to myocardial fibrosis by upregulating profibrotic cytokines and growth factors, such as transforming growth factor-beta (TGF-β) and platelet-derived growth factor (PDGF). This promotes the excessive deposition of extracellular matrix proteins, leading to stiffening of the cardiac tissue and impaired cardiac function.
== Mechanism of action == When a tumor stimulates the growth of new vessels, it is said to have undergone an 'angiogenic switch'. The principal stimulus for this angiogenic switch appears to be oxygen deprivation, although other stimuli such as inflammation, oncogenic mutations and mechanical stress may also play a role. The angiogenic switch leads to tumor expression of pro-angiogenic factors and increased tumor vascularization. Specifically, tumor cells release various pro-angiogenic paracrine factors (including angiogenin, vascular endothelial growth factor (VEGF), fibroblast growth factor (FGF), and transforming growth factor-β (TGF-β). These stimulate endothelial cell proliferation, migration and invasion resulting in new vascular structures sprouting from nearby blood vessels. Cell adhesion molecules, such as integrins, are critical to the attachment and migration of endothelial cells to the extracellular matrix.

Ipamorelin is a synthetic pentapeptide that stimulates growth hormone release by activating the ghrelin receptor. It is handled as a research tool rather than as a licensed therapeutic product. Its short chain length makes it comparatively simple to synthesize and analyze.
Both bind the same receptor, but ghrelin is a 28-amino-acid hormone with roles in appetite and energy balance. Ipamorelin is a much smaller synthetic peptide selected for growth hormone release with limited effect on appetite signaling in animal models. The full overlap between their effects is not completely mapped.
Most published data come from animal studies and a small number of human trials. Findings on growth hormone release are reasonably consistent, while long-term effects and direct comparisons with other secretagogues remain incompletely studied. Open questions include duration of action across different populations.
It has not been approved as a therapeutic by major regulators, and the human trial record is small and dated. Material available today is mostly sold as a research chemical for laboratory use. Approval and restriction status varies by country.
